Drug intelligence / Profile preview
THBS2 inhibitor refers to a class of therapeutic agents designed to target and inhibit Thrombospondin-2 (THBS2), a matricellular glycoprotein that is highly upregulated in fibrotic livers and plays a key role in driving liver fibrogenesis. Researchers at Beijing Friendship Hospital, Capital Medical University, identified a novel small molecule THBS2 inhibitor through integrated virtual drug screening and surface plasmon resonance (SPR) validation. In preclinical mouse models of liver fibrosis (induced by CCl4 or DDC), this inhibitor demonstrated potent anti-fibrotic efficacy and significant histopathological improvements. Mechanistically, THBS2 interacts with SPP1 and TLR4 to activate hepatic stellate cells (HSCs), and targeting the THBS2-SPP1 axis represents a promising synergistic therapeutic strategy for liver fibrosis.
