The-0504 is a novel, tumor protease-activatable nanotherapeutic designed to target the ferritin receptor (CD71/Transferrin receptor 1). It consists of a human ferritin (HFt)-based drug carrier conjugated to Genz-644282, a potent, wide-spectrum topoisomerase I inhibitor. The-0504 is engineered to remain inactive in healthy tissues and selectively release its payload within the tumor microenvironment upon activation by tumor-associated proteases. This targeted approach aims to overcome the systemic toxicity typically associated with CD71-targeting therapies and topoisomerase inhibitors. Preclinical studies have demonstrated high activity and low toxicity in models of aggressive human tumors, including breast and gastrointestinal cancers.
