THE002 is a preclinical-stage thermosensitive liposomal (TSL) formulation of irinotecan developed by Thermosome. The drug utilizes a proprietary lipid composition that remains stable at physiological temperatures but undergoes a phase transition to release its payload rapidly when exposed to mild local hyperthermia (typically 40-42°C). This approach is designed to maximize the concentration of irinotecan within the tumor microenvironment while minimizing systemic exposure and associated toxicities. Irinotecan, the active payload, is a camptothecin derivative that inhibits DNA topoisomerase I, leading to DNA strand breaks and cell death. THE002 is currently being investigated for the treatment of solid tumors, specifically pancreatic and colorectal cancers.
