**THEO-260** is a mechanistically differentiated **oncolytic adenovirus immunotherapy** designed to selectively lyse both cancer cells and cancer-associated fibroblasts (CAFs) in stromal-rich tumour microenvironments. By directly killing both tumour and CAF populations—while sparing normal fibroblasts and T cells—THEO-260 uniquely alleviates immune suppression and promotes immunogenic cell death, thereby activating effector T cell responses. It has shown efficacy in preclinical models and patient-derived ex vivo samples for ovarian cancer, including platinum-resistant disease, and demonstrates activity in other solid tumours (pancreatic, colorectal, lung, breast). The therapy is optimized for intravenous (IV) administration, with plans for intraperitoneal routes, and is stable under genetic and temperature stress. Developed by Theolytics, THEO-260 is currently being evaluated in a Phase I/IIa clinical trial in advanced-stage platinum-resistant ovarian cancer. It is produced via GMP manufacturing with high yields[1][2][3][4][5][6].
