Overview

Therapeutic autologous lymphocytes (NCI) for metastatic melanoma is an investigational adoptive cell therapy developed by the National Cancer Institute. The treatment involves isolating a patient's own CD4+ and CD8+ T lymphocytes, which are then expanded and sensitized ex vivo to recognize specific epitopes of the NY-ESO-1 cancer-testis antigen. NY-ESO-1 is frequently expressed in metastatic melanomas but not in normal adult tissues, except for the testis. Following a nonmyeloablative, lymphodepleting chemotherapy regimen (typically cyclophosphamide and fludarabine), these sensitized lymphocytes are reinfused into the patient. The therapy is often administered in conjunction with an NY-ESO-1 peptide vaccine and interleukin-2 (IL-2) to promote the survival and anti-tumor activity of the infused cells.
