therapeutic cytotoxic T lymphocytes-Fred Hutchinson Cancer Center-metastatic melanoma refers to an investigational adoptive cell therapy regimen developed by the Fred Hutchinson Cancer Center for the treatment of metastatic melanoma. This therapeutic approach involves the infusion of autologous cytotoxic T lymphocytes (CTLs) that have been expanded ex vivo and primed with interleukin-21 (IL-21) to enhance their anti-tumor potency and long-term persistence. The regimen is administered in combination with the CTLA-4 checkpoint inhibitor ipilimumab to further stimulate the immune response, along with lymphodepleting cyclophosphamide and supportive low-dose aldesleukin (IL-2). By combining tumor-specific T-cell transfer with checkpoint blockade, the therapy aims to overcome the immunosuppressive tumor microenvironment and achieve durable clinical responses in patients with advanced melanoma.
