The therapeutic double-plasmid hepatitis B virus deoxyribonucleic acid vaccine is a DNA vaccine designed to treat chronic Hepatitis B (CHB) infection. It comprises two non-infectious DNA plasmids: one encoding the Hepatitis B virus (HBV) core protein (Core) and the other encoding HBV polymerase (Pol). These plasmids are delivered into host cells, often via electroporation, leading to the endogenous production, processing, and secretion of the encoded Core and Pol proteins. These viral antigens are then presented in the context of MHC class I and II molecules, which in turn induces a robust cell-mediated immune response, specifically HBV-specific CD4+ and CD8+ T-cell responses. The primary goal of this vaccine is to induce a strong Th1 response to overcome the state of immune tolerance characteristic of chronic HBV infection.
