Theratope STn-KLH vaccine is an investigational therapeutic cancer vaccine developed as an active specific immunotherapy. It consists of a synthetic sialyl-Tn (STn) carbohydrate antigen conjugated to keyhole limpet hemocyanin (KLH), a high molecular weight protein carrier. The STn antigen mimics a tumor-associated carbohydrate found on the MUC1 mucin of many human adenocarcinoma cells, including breast, ovarian, colorectal, gastric, and pancreatic cancers. The mechanism involves stimulating both humoral and cellular immune responses against the tumor-associated antigen to inhibit tumor growth and potentially improve survival outcomes in patients with advanced cancers. Clinical development focused primarily on metastatic breast cancer; while the vaccine was well-tolerated with minimal toxicity (mainly local injection site reactions), pivotal phase III trials did not meet their primary efficacy endpoints[1][2][6][8].
