ThermoDox is a heat-activated liposomal formulation of the chemotherapeutic agent doxorubicin, originally developed by Celsion Corporation (now Imunon) using technology licensed from Duke University. The formulation is designed to remain stable in the bloodstream at normal body temperature but rapidly release its doxorubicin payload when exposed to hyperthermic temperatures (typically 40-42°C). This triggered release is intended to be used in conjunction with thermal ablation techniques, such as radiofrequency ablation (RFA), to deliver high concentrations of the drug directly to the margins of a tumor. While it was primarily investigated in a Phase 3 trial (the HEAT study) for hepatocellular carcinoma, the study failed to meet its primary endpoint. It is also being explored for other indications, including bladder cancer, through collaborations with the National Institutes of Health (NIH).
