Overview

Thiamine pyrophosphate (TPP), also known as thiamine diphosphate or cocarboxylase, is the active coenzyme form of vitamin B1 (thiamine). It is formed in the body by phosphorylation of thiamine and serves as an essential cofactor for several enzymes involved in carbohydrate metabolism, including pyruvate dehydrogenase and transketolase. TPP plays a critical role in decarboxylation reactions and the pentose phosphate pathway. Deficiency leads to impaired energy metabolism and neurological symptoms. Thiamine pyrophosphate is not typically administered directly as a drug; instead, thiamine or its salts are given, which are then converted to TPP in vivo[3][8][9].
