Thioplatin is a novel sulfur-containing, small molecule platinum-based chemotherapy agent. Originally developed by researchers at the German Cancer Research Center in Heidelberg and later advanced through agreements involving Antisoma and Waypharm, the drug is chemically identified as bis(O-ethyldithiocarbonato)platinum(II). It was specifically designed to exhibit enhanced antitumoral activity in the slightly acidic microenvironments typically surrounding tumors, a feature that distinguishes it from traditional platinum agents. Its mechanism of action involves binding to DNA to exert cytotoxic effects. Preclinical evaluations have demonstrated efficacy against small-cell lung cancer and colorectal cancer xenotransplants, as well as activity in cisplatin-resistant ovarian carcinoma cell lines.
