Overview

Thioureidobutyronitrile (Kevetrin) is a water-soluble, small-molecule alkylating agent that acts as an activator of the tumor suppressor protein p53. It is being developed primarily as an antineoplastic (anticancer) agent. The drug’s mechanism involves stimulating apoptosis, activating caspase 3, modulating poly(ADP-ribose) polymerase and retinoblastoma protein, and directly stimulating p53 activity. Thioureidobutyronitrile has received orphan drug status for several rare cancers including retinoblastoma, pancreatic cancer, and ovarian cancer. Its highest development phase reported is Phase II for ovarian cancer[1].
