Drug intelligence / Profile preview
Perchlozon (thioureidoiminomethylpyridinium perchlorate) is an oral small molecule anti-tuberculosis drug developed by Pharmasyntez in collaboration with the A.E. Favorsky Irkutsk Institute of Chemistry and the Saint-Petersburg Research Institute of Phthisiopulmonology. Approved in the Russian Federation in 2012, Perchlozon is indicated for the treatment of pulmonary tuberculosis, including multidrug-resistant (MDR-TB) and extensively drug-resistant (XDR-TB) strains. Chemically classified as a thiosemicarbazone, Perchlozon acts as a prodrug that requires bioactivation by the mycobacterial FAD-containing monooxygenase EthA. Once activated, it targets and inhibits the FAS-II dehydratase complex HadABC (comprising HadA, HadB, and HadC subunits), thereby disrupting mycolic acid biosynthesis and compromising the integrity of the mycobacterial cell wall.
