**tHIVconsvX vaccine** is an investigational second-generation conserved-region mosaic T-cell vaccine for HIV-1 prevention and therapeutic HIV-1 remission strategies. It encodes bivalent mosaic immunogens derived from six functionally conserved HIV-1 Group M regions, including two Gag regions and four Pol regions, to broaden coverage of globally circulating HIV-1 variants. In clinical studies, the immunogens have been delivered using non-replicating ChAdOx1 chimpanzee adenovirus and Modified Vaccinia Ankara vectors in a prime-boost regimen, with ChAdOx1.tHIVconsv1 followed by MVA.tHIVconsv3 and MVA.tHIVconsv4. The intended effect is induction of broad HIV-1-specific cytotoxic T-cell responses against conserved Gag and Pol epitopes, aiming to suppress viral replication and support antiretroviral therapy-free control.
