THN201 is an investigational fixed-dose combination therapy developed by Theranexus for the treatment of cognitive disorders associated with Alzheimer's disease. The drug combines donepezil, a standard-of-care acetylcholinesterase inhibitor, with mefloquine, a small molecule repurposed to act as a blocker of glial connexin channels (specifically Connexin 30 and Connexin 43). The therapeutic rationale is based on the Theranexus platform, which suggests that modulating glial gap junction communication can enhance the pro-cognitive effects of neuronal-targeting drugs like donepezil. By inhibiting these connexins, THN201 aims to improve synaptic transmission and neuronal-glial interactions, potentially offering superior efficacy over donepezil monotherapy. The development of THN201 has been supported by the French government's Cx-COG project in collaboration with academic partners and the CDMO Synerlab Développement.
