Thorium-227 anetumab corixetan is a novel radiopharmaceutical designed as a mesothelin-targeted thorium conjugate (MSLN-TTC) for the treatment of solid tumors that overexpress the mesothelin protein. It consists of a fully human monoclonal antibody (anetumab) specific for mesothelin, linked via an octadentate 3,2-HOPO chelator to the alpha-emitting radionuclide thorium‑227. This structure enables highly selective delivery of potent alpha radiation directly to tumor cells expressing mesothelin, minimizing damage to healthy tissues. The drug is being developed primarily for ovarian cancer but has potential applications in other cancers such as malignant pleural and peritoneal mesothelioma and pancreatic ductal adenocarcinoma. Its mechanism involves binding to tumor cells via the antibody component, followed by localized emission of cytotoxic alpha particles from thorium‑227 decay, resulting in DNA damage and cell death[1][4][6].
