Overview

THR-317 is a recombinant humanized monoclonal antibody that targets and inhibits placental growth factor (PlGF). PlGF is a protein involved in pathological angiogenesis and vascular leakage. By blocking PlGF at its receptor-binding site, THR-317 aims to reduce abnormal blood vessel growth and leakage in the retina. The drug was primarily developed for ophthalmic indications such as diabetic macular edema (DME) and diabetic retinopathy (DR), with additional exploration in oncology indications like medulloblastoma and glioblastoma. In preclinical models and clinical studies, THR-317 demonstrated anti-inflammatory as well as anti-edema properties. It has been evaluated both as monotherapy and in combination with standard-of-care anti-VEGF agents such as ranibizumab (Lucentis). Clinical trials showed that intravitreal administration of THR-317 was safe and well tolerated; efficacy signals were observed particularly among patients who responded poorly to prior anti-VEGF therapy[1][2][4][5][6][7][8].
