Overview

Thrombomodulin alfa is a recombinant, soluble form of human thrombomodulin comprising all extracellular domains of the native protein. It acts as an anticoagulant by binding to thrombin, which then activates protein C. Activated protein C inactivates factors Va and VIIIa in the presence of cofactor protein S, thereby inhibiting further thrombin generation and clot extension. Thrombomodulin alfa also exhibits anti-inflammatory effects by activating thrombin–activatable fibrinolysis inhibitor and inhibiting neutrophil migration. It is primarily developed for the treatment of disseminated intravascular coagulation (DIC) and other blood clotting disorders such as thromboembolism[1][2][3][4][5].
