Overview

This is a combination therapy consisting of a thrombopoietin mimetic (such as eltrombopag, romiplostim, or recombinant human thrombopoietin) and cyclosporin A. Thrombopoietin mimetics are agents that stimulate the thrombopoietin receptor (MPL), leading to increased proliferation and differentiation of megakaryocyte progenitors and enhanced platelet production. Cyclosporin A is an immunosuppressive agent that inhibits calcineurin, thereby suppressing T-cell activation. The combination has been investigated primarily for the treatment of aplastic anemia and corticosteroid-resistant immune thrombocytopenia (ITP), where it aims to restore hematopoiesis by both stimulating platelet production and modulating immune-mediated marrow suppression[2][4][5]. Clinical studies have shown improved response rates in refractory ITP and aplastic anemia with this regimen compared to monotherapy[2][3][4].
