TSP-1 shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of thrombospondin-1 (TSP-1), also known as THBS1. In the context of glioblastoma, TSP-1 has been identified as a synaptogenic factor secreted by tumor cells that promotes the formation of excitatory synapses between neurons and glioma cells. This synaptic integration induces local hyperexcitability and drives tumor proliferation and tumor microtube (TMT) formation. By utilizing RNA interference to knock down TSP-1, this agent aims to disrupt glioma-induced network remodeling and inhibit tumor expansion. Research indicates that TSP-1 mediates these effects through the gabapentin α2δ-1 receptor, and silencing TSP-1 has been shown to reduce tumor growth and improve survival in preclinical glioblastoma models.
