**THRV-1257** is a selective small molecule inhibitor of serum- and glucocorticoid-inducible kinase 1 (SGK1) developed by Thryv Therapeutics for advanced solid tumors, particularly anaplastic thyroid cancer (ATC). It targets SGK1, a kinase downstream of PI3K/AKT signaling that promotes tumor growth and immune evasion, showing promising preclinical efficacy including tumor growth inhibition alone and enhanced effects in combination with standard-of-care therapies in BRAFV600E-mutant models.
