Overview

THRV-1268 is a novel, potent small molecule inhibitor of serum/glucocorticoid-regulated kinase 1 (SGK1), developed by Thryv Therapeutics. SGK1 is a PI3-kinase-dependent serine/threonine-protein kinase involved in metabolic signaling pathways that are crucial in cardiometabolic diseases. Inhibition of SGK1 has been shown to reduce QTc interval prolongation, arrhythmias, fibrosis, and heart failure in preclinical models. THRV-1268 is being developed primarily for the treatment of atrial fibrillation and heart failure with preserved ejection fraction (HFpEF). Preclinical studies have demonstrated its efficacy in reducing the development of heart failure and fibrosis as well as mitigating biomarkers associated with adverse cardiovascular changes. The drug is currently undergoing Phase I clinical trials for paroxysmal atrial fibrillation and diastolic heart failure[1][3][4][6][7].
