Drug intelligence / Profile preview
THX-B is a potent, non-peptidic small molecule antagonist of the p75 neurotrophin receptor (p75NTR, also known as NGFR or CD271). It acts as a competitive blocker of pro-nerve growth factor (proNGF) binding to p75NTR, thereby inhibiting both proNGF-induced and ligand-independent p75NTR signaling pathways. Developed by researchers at McGill University, THX-B has demonstrated therapeutic potential in preclinical models for several conditions, including diabetic retinopathy, diabetic voiding dysfunction, age-related bladder remodeling, retinitis pigmentosa, and metastatic melanoma. By blocking p75NTR, THX-B reduces neuroinflammation, prevents neuronal apoptosis (such as retinal ganglion cell death), and limits tumor lymphangiogenesis and distal metastasis.
