Overview

Thymalfasin + toripalimab is an investigational combination immunotherapy regimen consisting of thymalfasin (Thymosin Alpha 1), a synthetic peptide immunomodulator, and toripalimab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Thymalfasin acts as an immunostimulant that enhances T-cell maturation and function, potentially counteracting immune senescence in older populations. Toripalimab is a checkpoint inhibitor that restores anti-tumor T-cell activity by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2). The combination is primarily being evaluated in Phase 2 clinical trials sponsored by Sun Yat-sen University for the treatment of advanced, unresectable, or metastatic melanoma in elderly patients (aged 60 and older), where it aims to leverage the immunomodulatory properties of thymalfasin to enhance the efficacy of PD-1 blockade.
