Thymoproteasome-expressing mesenchymal stromal cells (MSC-TPr) are an experimental cell-based immunotherapy and cellular vaccine developed by CELL Technologies and IntelliStem Technologies. These mesenchymal stromal cells (MSCs) are genetically engineered to express the thymoproteasome (TPr) complex, a specialized proteasome typically found in cortical thymic epithelial cells that is essential for the positive selection of CD8+ T-cells. In a cancer context, MSC-TPr acts as an immune-modifying agent that, upon administration, recruits myeloid cells—specifically dendritic cells and macrophages. The dendritic cells cross-prime endogenous T-cells with tumor antigens, triggering a potent and specific CD8+ T-cell-mediated anti-tumor immune response. Preclinical studies in lymphoma and melanoma models have demonstrated that this approach can lead to complete protection and tumor regression, particularly when inhibitory macrophage populations are managed.
