Overview

Thymosin fraction 5 (TF5) is a partially purified bovine thymic extract containing a mixture of biologically active polypeptides, including thymosin alpha-1 and thymosin beta-4. It was one of the first thymic hormone preparations to be extensively studied for its immunomodulatory properties. TF5 works by promoting the differentiation and functional maturation of T-lymphocytes from precursor cells and enhancing various immune functions. In clinical research, it has been explored as an adjuvant therapy for various cancers and primary immunodeficiency diseases. Notably, TF5 has been shown to induce the expression of pyrimidine nucleoside phosphorylase enzymes, such as uridine phosphorylase, which can potentiate the activity of fluoropyrimidine chemotherapy prodrugs like 5'-deoxy-5-fluorouridine by increasing their conversion to active 5-fluorouracil.
