Thymus nuclear extract is a biologic preparation derived from the nuclei of bovine (calf) thymus cells. It is a complex mixture containing various soluble nuclear antigens (SNAs), most notably Sm, nRNP, Ro (SS-A), La (SS-B), Scl-70, and Jo-1. Historically, its primary use has been as a diagnostic reagent in clinical immunology, serving as the substrate for double immunodiffusion (Ouchterlony), ELISA, and hemagglutination assays to detect antinuclear antibodies (ANAs) in patients with systemic autoimmune rheumatic diseases (SARDs) such as systemic lupus erythematosus (SLE), mixed connective tissue disease (MCTD), and polymyositis. Beyond its diagnostic utility, bovine thymus nuclear extract has been investigated in clinical trials (e.g., NCT06795945) to evaluate its safety, tolerability, and potential immunomodulatory effects in healthy adults, suggesting potential applications as a therapeutic agent or dietary supplement for immune system support.
