TI-0032-III is an investigational in vivo CAR-T cell therapy developed by Therorna for the treatment of relapsed and refractory B cell-mediated autoimmune diseases. The therapeutic product consists of lipid nanoparticles (LNPs) that target T cells and encapsulate circular RNA (circRNA) encoding a CD19-specific chimeric antigen receptor (CAR). Unlike traditional ex vivo CAR-T therapies that require complex cell manufacturing outside the body, TI-0032-III is designed to reprogram T cells directly within the patient's body to identify and deplete pathogenic B cells. This approach aims to provide a more scalable and accessible treatment for various autoimmune conditions, including systemic lupus erythematosus (SLE), Sjögren's syndrome, and systemic sclerosis.
