Ti-061

Ti-061 is an investigational humanized IgG4 monoclonal antibody targeting CD47, a cell-surface protein that acts as a "don't eat me" signal to the immune system. By binding to CD47, Ti-061 blocks its interaction with SIRPα on macrophages, thereby promoting the phagocytosis of cancer cells. Developed by Tioma Therapeutics (later Arch Oncology), the drug was designed as an innate immune checkpoint inhibitor for the treatment of advanced solid tumors. Clinical development included a Phase 1/2 study in the United Kingdom evaluating Ti-061 as both a monotherapy and in combination with the PD-1 inhibitor pembrolizumab; however, the trial was prematurely terminated in 2017.
