TI-235 is an autologous T-cell receptor (TCR) engineered regulatory T-cell (Treg) therapy being developed by TeraImmune for the treatment of multiple sclerosis (MS). The therapy utilizes the company's proprietary Treg-Amp platform to expand stable regulatory T cells, which are then engineered to express a TCR specific for myelin basic protein (MBP), a primary autoantigen in the pathogenesis of MS. By targeting MBP, these specialized Tregs are designed to localize to the central nervous system and suppress the localized neuroinflammation and autoimmune attack on the myelin sheath. TI-235 has demonstrated efficacy in preclinical models of experimental autoimmune encephalomyelitis (EAE) and is currently in Early Phase 1 clinical development.
