tiacrilast

Tiacrilast is a small molecule mast cell stabilizer originally developed by Hoffmann-La Roche for the treatment of allergic and inflammatory conditions, including atopic dermatitis (eczema) and asthma. It functions by inhibiting the degranulation of mast cells, thereby preventing the release of histamine and other inflammatory mediators. In clinical evaluations, a 3% hydrogel formulation was tested for atopic eczema; however, it failed to demonstrate a statistically significant clinical benefit over the placebo vehicle, leading to the conclusion that mast cell degranulation may not be the primary driver in the maintenance of chronic eczema lesions. Consequently, development for this indication was discontinued.
