Tiadenol is a hypolipidemic agent and a peroxisome proliferator used for the treatment of hypercholesterolemia and hypertriglyceridemia. It belongs to a class of compounds that induce hepatic peroxisome proliferation in rodents, similar to fibrates. Tiadenol acts by modulating the metabolism of fatty acids and phospholipids, specifically inducing peroxisomal beta-oxidation, stearoyl-CoA desaturase, and various acyltransferases. It is metabolized into thiodicarboxylic acids, such as bis(carboxymethylthio)-1,10-decane, which are considered the active metabolites responsible for its enzymatic induction effects. Clinical studies have demonstrated its efficacy in reducing total cholesterol, VLDL, and LDL while increasing HDL levels. It was historically developed by Midy and has been marketed in several European countries.
