Overview

Tiamulin fumarate is a semi-synthetic pleuromutilin antibiotic primarily utilized in veterinary medicine for the treatment of swine dysentery caused by Brachyspira hyodysenteriae and mycoplasmal pneumonia. It exerts its antimicrobial effect by binding to the peptidyl transferase center of the bacterial 50S ribosomal subunit, thereby inhibiting protein synthesis. Beyond its established antibiotic properties, recent high-throughput screening has identified tiamulin fumarate as a potential small-molecule inhibitor of Tumor Necrosis Factor-alpha (TNF-α). In preclinical models of psoriasis, it has demonstrated the ability to block NF-κB and MAPK signaling pathways, leading to improved dermatitis symptoms. This dual activity suggests potential for drug repositioning in human inflammatory conditions.
