TicTwo Therapeutics is developing a series of brain-penetrant, allosteric small molecule inhibitors of Tyrosine Kinase 2 (TYK2) for the treatment of Alzheimer's Disease and Frontotemporal Dementia (FTD). These compounds are specifically engineered to cross the blood-brain barrier, a feature that distinguishes them from existing TYK2 inhibitors like deucravacitinib. The therapeutic approach focuses on mitigating excessive neuroinflammation driven by interferon signaling, which is implicated in the pathology of neurodegenerative diseases, particularly those involving TDP-43. By binding to the allosteric pseudokinase domain of TYK2, these molecules aim to provide high selectivity and reduce the risk of off-target effects associated with traditional JAK inhibitors. The program is currently in the preclinical stage, supported by genetic data linking TYK2 loss-of-function to neuroprotection.
