Drug intelligence / Profile preview
A **Tie2 activating antibody** (or Tie2-agonistic antibody) is a class of therapeutic monoclonal antibodies designed to bind and activate the endothelial receptor tyrosine kinase Tie2 (also known as TEK). Tie2 signaling is crucial for maintaining vascular stability, promoting endothelial cell survival, and reducing vascular permeability and inflammation. Under pathological conditions such as chronic kidney disease (CKD), sepsis, diabetic retinopathy, and cancer, Tie2 signaling is often impaired due to the downregulation of its agonist angiopoietin-1 (ANGPT1) and the upregulation of its antagonist angiopoietin-2 (ANGPT2). Tie2 activating antibodies, such as PMC-403, ABTAA, and 4E2, restore Tie2 phosphorylation and downstream Akt/ERK signaling. This leads to vascular normalization, tightening of endothelial junctions, and reduction of capillary rarefaction and tissue fibrosis. Clinical development is currently led by candidates like PharmAbcine's PMC-403, which is in Phase 1 clinical trials for neovascular age-related macular degeneration (nAMD).
