Tie2-Ang2 blocking antibodies are a class of therapeutic monoclonal antibodies designed to disrupt the interaction between the receptor tyrosine kinase Tie2 (TEK) and its ligand angiopoietin-2 (Ang2/ANGPT2). Tie2 is primarily expressed on endothelial cells and tumor-associated macrophages, where it regulates vascular stability, angiogenesis, and inflammation. Under pathological conditions such as cancer, elevated levels of Ang2 act as a context-dependent antagonist or weak agonist of Tie2, destabilizing blood vessels and promoting tumor angiogenesis, vascular permeability, and metastasis. By blocking the Tie2-Ang2 signaling axis, these antibodies promote vascular normalization, reduce tumor cell intravasation and dissemination (particularly through tumor microenvironment of metastasis, or TMEM, doorways), and inhibit tumor growth. This therapeutic approach is being investigated for the treatment of various solid tumors, including breast cancer, as well as retinal vascular diseases.
