Tigapotide is a synthetic 15-amino acid peptide derived from amino acids 31–45 of the human prostate secretory protein PSP94, one of the three predominant proteins in seminal fluid. It was developed as an investigational anticancer agent, primarily for hormone-refractory prostate cancer. Tigapotide exerts its effects by inhibiting matrix metalloproteinase-9 (MMP-9) secretion and its interaction with cell surface receptor CD44, interfering with vascular endothelial growth factor (VEGF) signaling pathways to produce antiangiogenic effects, and reducing parathyroid hormone-related protein (PTHrP) levels. These actions contribute to inhibition of tumor angiogenesis, metastasis, and potentially induce apoptosis in cancer cells. Preclinical studies also suggest activity against breast and colon cancer cells via laminin receptor-mediated mechanisms[1][2][3][4][6].
