Overview

Tigecycline + gentamicin is a combination of two antibiotics used primarily for the treatment of severe, multidrug-resistant bacterial infections. Tigecycline is a glycylcycline antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, blocking entry of amino-acyl tRNA molecules and preventing peptide chain elongation. Gentamicin is an aminoglycoside antibiotic that binds to the 30S ribosomal subunit as well, causing misreading of mRNA and inhibiting protein synthesis, leading to bactericidal activity. The combination has been studied for synergistic effects against difficult-to-treat pathogens such as carbapenem-resistant Klebsiella pneumoniae (CRKP) and Enterococcus faecium in both in vitro and in vivo models. While synergy rates vary depending on pathogen and study design, this combination can provide enhanced efficacy compared to monotherapy in certain clinical scenarios[1][2][3][5]. Both drugs are administered intravenously.
