Tigliptin is an orally active, small molecule dipeptidyl peptidase-4 (DPP-4) inhibitor developed by the Tianjin Institute of Pharmaceutical Research (TIPR) for the treatment of type 2 diabetes mellitus. As a member of the gliptin class, tigliptin works by selectively inhibiting the DPP-4 enzyme, which is responsible for the rapid degradation of incretin hormones such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). By stabilizing these hormones, tigliptin enhances glucose-dependent insulin secretion from pancreatic beta cells and suppresses glucagon release from alpha cells, thereby improving glycemic control with a low risk of hypoglycemia. The drug has progressed into Phase 3 clinical trials in China, where it is being evaluated for its efficacy and safety as a monotherapy and in combination with other antidiabetic agents.
