Overview

TIK-101 is an investigational, once-daily, extended-release oral formulation of baclofen, a selective gamma-aminobutyric acid (GABA) B receptor agonist. Developed by Tikvah Therapeutics using Depomed's proprietary AcuForm gastric-retentive drug delivery technology, the formulation was designed to provide consistent therapeutic levels of baclofen over a 24-hour period. This approach aimed to improve patient compliance and minimize the side effects—such as sedation, somnolence, and muscle weakness—typically associated with the peak-and-trough plasma concentrations of standard immediate-release baclofen. The primary clinical focus for TIK-101 was the management of spasticity in patients with multiple sclerosis or spinal cord injury. Although the drug candidate reached Phase 3 clinical trials, development was terminated in 2010 following the dissolution of Tikvah Therapeutics.
