**TIL-NPs** are investigational multifunctional PEG-PLGA nanoparticles designed to activate and expand tumor-infiltrating lymphocytes directly within tumors. The nanoparticles are surface-conjugated with anti-CD3 antibody, anti-4-1BB antibody, and interleukin-2, enabling coordinated T-cell receptor stimulation, 4-1BB co-stimulation, and cytokine support. Developed at the University of Texas Southwestern Medical Center, intratumoral TIL-NPs produced local tumor regression and systemic antitumor immune responses in preclinical colorectal cancer and humanized renal cell carcinoma models, including activity against contralateral tumors and generation of memory T-cell responses. The program remains preclinical, with no reported clinical trials or marketed product.
