Tilavonemab is a humanized IgG4 monoclonal antibody developed as an immunotherapy targeting tauopathies such as Alzheimer's disease and progressive supranuclear palsy. It binds to the N-terminal region of aggregated, extracellular pathological tau protein, which is implicated in the propagation of neurofibrillary pathology in these diseases. By binding to soluble extracellular tau, tilavonemab aims to block its cell-to-cell spread and reduce the progression of tau pathology in the brain[2][3][4]. The drug was originally developed by C2N Diagnostics and AbbVie[7]. Clinical trials showed that while tilavonemab was generally well tolerated, it did not demonstrate significant efficacy compared with placebo for either Alzheimer's disease or progressive supranuclear palsy[3][5].
