Tilpisertib is a synthetic small molecule inhibitor of the serine/threonine kinase Cot, also known as MAP3K8 or TPL2. It was developed by Gilead Sciences as a potential anti-inflammatory drug, primarily targeting auto-inflammatory diseases such as ulcerative colitis. Tilpisertib acts upstream in the MEK-ERK pathway and is expected to reduce production and signaling of tumor necrosis factor alpha (TNFα), thereby modulating inflammatory responses. The compound has been evaluated in phase 2 clinical trials for moderately to severely active ulcerative colitis but development appears to have been discontinued for this indication[1][3][5].
