Overview

Tilrekimig is a trispecific engineered IgG1 monoclonal antibody that simultaneously targets human interleukin 4 (IL-4), interleukin 13 (IL-13), and thymic stromal lymphopoietin (TSLP), key cytokines driving type 2 inflammation in atopic dermatitis and related allergic and inflammatory diseases.[1][3][5][13][14] It is constructed as a complex fusion protein in which anti‑IL‑13, anti‑IL‑4, and anti‑TSLP binding domains derived from humanized mouse monoclonal antibodies are connected via engineered peptide linkers and Fc modifications that minimize effector function and extend half‑life.[5] Developed by Pfizer, tilrekimig is intended as a single biologic that broadly suppresses type 2 inflammatory signaling upstream in the pathway, with a therapeutic claim focused on treatment of atopic dermatitis and other immune and inflammatory conditions.[5][13]
