Drug intelligence / Profile preview
This experimental cell therapy, developed by the Second Affiliated Hospital of Guangzhou Medical University, utilizes autologous tumor-infiltrating lymphocytes (TILs) and CAR-engineered TILs (CAR-TILs) for the treatment of advanced solid tumors. The therapy employs CRISPR-Cas9 technology to knockout the PD-1 gene (PDCD1) in the harvested lymphocytes, aiming to prevent T-cell exhaustion and enhance anti-tumor persistence. Furthermore, the cells are genetically modified to secrete single-chain variable fragments (scFvs) targeting PD-1 and CTLA-4, providing localized checkpoint inhibition within the tumor microenvironment. Depending on the specific patient and tumor type, the cells may also express Chimeric Antigen Receptors (CARs) targeting a variety of tumor-associated antigens, including HER2, Mesothelin, EGFR, and Claudin 18.2. This multi-mechanistic approach is currently being evaluated in a Phase 1 clinical trial to assess safety and preliminary efficacy.
