Overview

TILT-321 is a preclinical **oncolytic adenovirus** immunotherapy from TILT Biotherapeutics for solid tumors. It is an Ad5/3 chimeric, tumor-selective adenovirus engineered to encode a **bispecific T-cell engager** that binds **Mucin 1** on tumor cells and **CD3** on T cells, thereby promoting local T-cell activation and redirected cytotoxicity within the tumor microenvironment. In published preclinical work, TILT-321 infected tumor cells secreted a functional anti-MUC1 anti-CD3 engager, increased intratumoral T-cell activity, and produced antitumor effects in humanized xenograft models, supporting its use as a locally produced virus-delivered immunotherapy intended to overcome systemic exposure and short half-life limitations of recombinant BiTE-like molecules in solid tumors.
