Overview

TILT-322 is a novel chimeric **oncolytic adenovirus** engineered to deliver targeted immunotherapy against solid tumors, particularly ovarian cancer. The construct utilizes an Ad5/3-E2F-d24 backbone with selective tumor replication, featuring a bispecific T cell engager targeting Mucin 1 (MUC1), a tumor-associated antigen, and CD3 on T cells, thereby stimulating potent anti-tumor T cell cytotoxicity. Additionally, it expresses human interleukin-2 (IL-2) to further activate and expand effector T cells. The agent is designed to both lyse tumor cells directly and modulate the tumor microenvironment by enhancing immune cell infiltration and reversing T cell exhaustion, offering promising anti-tumor efficacy in preclinical and translational models. The leading developer is TILT Biotherapeutics, a company focused on next-generation immuno-oncology therapies[1][4][7][8].
