Overview

TILT-452 is a **preclinical cytokine-armed oncolytic adenovirus** being developed by TILT Biotherapeutics for solid tumors. The construct is described as an Ad5/3 chimeric adenovirus with tumor-selective replication features and encodes a **variant human interleukin-2** designed to preferentially stimulate IL-2 receptor signaling on effector lymphocytes over regulatory T cells. Mechanistically, it combines **direct oncolytic tumor cell killing** with **local immunostimulation**, aiming to enhance antitumor activity of tumor-infiltrating lymphocytes and natural killer cells in immunosuppressive solid-tumor microenvironments. Preclinical work has specifically highlighted ovarian cancer and pancreatic ductal adenocarcinoma models, and public pipeline summaries indicate the asset remains in preclinical development for solid tumors.
