Overview

TILT-517 is an investigational **oncolytic adenovirus therapy** developed by TILT Biotherapeutics for cancer immunotherapy, with reported development focus in **renal cell carcinoma**. It is a genetically modified adenovirus based on an Ad5/3 chimeric backbone and an E2F promoter with a d24 deletion designed to support selective replication in tumor cells, leading to **direct oncolysis** of malignant tissue. The virus is engineered to express **human interleukin-7**, with the goal of enhancing anti-tumor immunity by promoting activation and infiltration of immune effector populations such as CD4-positive T cells, CD8-positive T cells, natural killer cells, and natural killer T cells within the tumor microenvironment. Preclinical information indicates activity both as monotherapy and in combination with immune checkpoint blockade, including anti-PD-1 and anti-PD-L1 approaches, particularly in immunosuppressive tumor settings such as renal cell carcinoma.
