Drug intelligence / Profile preview
A dual-targeting cell therapy for Acute Myeloid Leukemia (AML) utilizing cytokine-induced killer (CIK) cells. The therapy is engineered using the "IF-BETTER" strategy, which pairs a second-generation chimeric antigen receptor (CAR) with a cytokine-costimulatory receptor (CCR) to simultaneously target CD33 and the leukemic stem cell (LSC) marker TIM-3. A distinguishing feature is the use of a TIM-3-specific single-chain fragment variable (scFv) that preferentially binds to AML-specific hyper-fucosylated and hyper-sialylated N-glycans, thereby enhancing selectivity for leukemic cells and reducing on-target/off-tumor toxicity against healthy TIM-3-expressing immune cells. Developed by researchers at the University of Milano-Bicocca and IRCCS San Gerardo dei Tintori, this approach aims to overcome disease heterogeneity and achieve durable leukemia clearance.
